MIT researchers have developed a two-dose HIV vaccine strategy that may be as effective as previous seven-dose regimens. The study, published in Science Immunology, shows that administering a smaller first dose followed by a larger second dose one week later generates stronger immune responses.

The vaccine uses an HIV envelope protein and a nanoparticle adjuvant and was tested using computational modeling and experiments in mice. Results indicate significant improvements in T-cell and antibody responses, making this approach more practical for mass vaccination campaigns. A single-dose version is already in clinical trials.
This research, led by MIT’s Arup Chakraborty and Darrell Irvine from the Scripps Research Institute, could help overcome HIV’s rapid mutation and potentially be adapted for other vaccines.
